does anyone know if they are making a film of Eldest, the second in the trilogy?

the movie Eragon apparently didn't do so well so Eldest might not happen but I don't know for sure

It's not a trilogy any more. In a newsletter I received, Christopher Paolini said the last book was going to be too long, so he split it in two. As for the Eldest movie, there's a rumor that it's coming out in 2011, though there's a poster claiming it comes out December 2009. The source isn't very reliable (Wikipedia), but that's all I could find about it.

Well, Paolini is calling is a "Cycle," but it could also be a "Quartet." A few months ago Christopher Paolini said that he doubts that there will be a filming of the second book, because to put it nicely, the first movie was a flop. Seriously. My library won't even buy one copy of it because of that. Also, from the first movie, they missed the part where he got the scar which is a huge part in the second book, so people that didn't read the books might get confused if they go on with that without it ever happening in the first movie. Overall, unless they re-do the first movie, no.

To answer your first question: They made a first movie, they'll likely make a second. And I really hope they do a better job of the second movie, I walked out halfway through the first because they did such a bad job. Didn't follow the book at all.
Second question: It is now a Quartet.
